MLPA is a simple method for relative quantification of up to 45 nucleic acid sequences in a single reaction. MLPA is: Sensitive: For MLPA analysis only 20 ng DNA or 10 ng total RNA are required. Specific: MLPA can be used to distinguish sequences differing in only one nucleotide. Reproducible: Coefficient of variability of each probe is 3% to 8 % when human DNA samples of 40-100 ng are analyzed. Simple: MLPA can be performed as a one tube reaction. MLPA is an addition-only method. High throughput: Processing of 96 samples will require less than three hours hands-on time. Low cost per data point: All reagents included, necessary equipment is present in most molecular biology laboratories.
