SALSA MLPA KIT P040 CLL

CHRONIC LYMPHOCYTIC LEUKEMIA (CLL) is the most frequent type of leukemia in the western world and results in the progressive accumulation of mature CD5(+) B lymphocytes in blood and bone marrow of the affected person. Chromosomal translocations are rare events in B-CLL. Copy number changes of certain chromosomal regions are however frequent. Some of these have been found to be highly prognostic markers of this disease. SALSA MLPA probemixes P037 and P038 contain probes for several genomic regions that are recurrently imbalanced in B cell CLL. On request of some customers we have prepared an alternative probemix P040, which contains a selection of probes in P037 and P038. This P040 CLL MLPA kit can be used to determine the loss of TP53 (17p13; 7 probes), the RB1 / DLEU / MIRN15A-16 region on 13q14 (10 probes), the ATM gene region on 11q23 (7 probes) as well the presence of trisomy 12 (9 probes) in DNA samples obtained from chronic lymphocytic leukemia. In addition, reference probes on various other chromosomes are present. For each region to be analysed, several probes are present. This might be necessary for correct analysis of samples containing lower (30-60%) percentages of tumor cells. Please note that these probemixes have not been used on CLL DNA samples at MRC-Holland. Suggestions for improvement of this P040 kits are appreciated.

This kit is not CE/FDA certified for use in diagnostic procedures. Salsa MLPA kits are supplied with all necessary buffers and enzymes. Purchase of the Salsa MLPA test kits includes a limited license to use these products for research purposes. The use of this MLPA kit requires a thermocycler with heated lid and sequence type electrophoresis equipment. Different fluorescent PCR primers are available. The MLPA technique has been first described in Nucleic Acid Research 30, e57 (2002)
