SALSA MLPA KIT P181-P182 Centromere

SALSA MLPA kits P181 & P182 can be used to detect changes in copy number of genes close to the centromeres of all chromosomes, with the exception of the Y-chromosome. In most cases, we designed probes for the first well-characterised gene near the centromeric region.
Both the P181 probemix and the P182 probemix contain one probe for each of the chromosome arms. Probes in P181 and P182 detect different sequences. Human chromosomes 13, 14, 15, 21 and 22 each have more than 10 Mb of repeat sequences at one end, covering most or all of the p arms. For these chromosomes, we have used two probes on the q arm, close to the centromere.
Possible applications of these probe mixes are in cancer research, as well as for characterisation of marker chromosomes.
This MLPA kit is designed to detect deletions/duplications of one or more genes near the centromeres. Deletions of probe recognition sequences will be apparent by a 35-50% reduced relative peak area of the amplification product of that probe. However, mutations/polymorphisms very close to the probe ligation site may also result in a reduced relative peak area. Apparent deletions of a single exon therefore always require confirmation by other methods. We have no information on what percentage of defects in these genes is caused by deletions/duplications of complete exons. Please note that most defects in these genes are expected to be small (point) mutations, most of which will not be detected by this MLPA test.
