SALSA MLPA KIT R011 Apoptosis mRNA
[
http://www.geneclinics.org/profiles/hht/details.html]
Lot 0108: Several probes have been added or replaced as compared to previous lots 1106 & 0105.
This SALSA MLPA R011 Apoptosis mRNA probe mix contains several probes specific for human mRNAs that are important for regulating the apoptosis pathways. The total probe mix contains 39 different probes, among which probes for BAX, BAK1, BCL2, BID, BCL2L1 and BBC3 (PUMA) mRNAs.
MLPA analysis of RNA requires a minimum of only 10 ng total RNA. Strong signals of probes specific for abundant mRNAs can be reduced by inclusion of a competitor oligonucleotide containing the same hybridizing sequence as the synthetic probe oligonucleotides, but lacking the PCR primer sequence. In the R011 mix, the probe signals of B2M, FTL, PARN, BCL2L1, BIRC6, BNIP3L and MCL1 (184 nt fragment) mRNAs are deliberately reduced by the inclusion of competitor oligonucleotides.
An MLPA reaction with the use of this probe mix on samples of human total RNA will result in a mixture of up to 39 amplification products that can be separated by capillary electrophoresis. The length of each amplification product indicates the probe from which it was derived. The relative amount of each amplification product reflects the relative amount of each probe target sequence in the sample analysed.

Current Lot Number.: Lot 0108
IMPORTANT NOTICE:
MLPA kits are sold by MRC-Holland for research purposes and to demonstrate the possibilities of the MLPA technique. This kit is not CE/FDA certified for use in diagnostic procedures. Salsa MLPA kits are supplied with all necessary buffers and enzymes. Purchase of the Salsa MLPA test kits includes a limited license to use these products for research purposes.
The use of this MLPA kit requires a thermocycler with heated lid and sequence type electrophoresis equipment. Different fluorescent PCR primers are available. The MLPA technique has been first described in Nucleic Acid Research 30, e57 (2002)
