15. When I received my order, all dry ice was gone. Will the MLPA kits be damaged?
More than 98% of all shipments arrive without any problems. Unfortunately mistakes at TNT or Fedex, or customs / FDA etc problems can cause delays of shipments. Custom clearance in some countries like India and Brazil can be very problematic. All items in MLPA kits are fortunately very stable. When testing product stability, we found that, after incubating all SALSA MLPA ingredients (probemix, buffers & enzymes) at room temperature for 11 weeks, good MLPA results were still obtained. However, if you obtain any problems, we guarantee a replacement.

1. How can I order from MRC-Holland?
Orders can be placed by e-mail: info@mlpa.com; by fax: +31-20-6891149; by mail: MRC-Holland, Hudsonstraat 68, 1057SN Amsterdam, The Netherlands, or through one of our distributors.
SALSA MLPA kits are supplied with all necessary buffers and enzymes. Our MLPA products are shipped worldwide directly from Amsterdam, the Netherlands on dry ice or with ice packs by courier service on the first Monday after we receive the order. Orders within Europe are shipped on Monday and Wednesday.

*PCR Primer Labels
For most ABI sequencing apparatus, we recommend FAM-labelled fluorescent PCR primers Several PCR primers with other fluorescent labels are available, including Cy5.0 (Beckman), ROX, and IRD800 (LICOR). Which label is ordered does not affect the price of a kit. Please mention on each order the required fluorescent label of the PCR primer.
**V.A.T. number (European Union only)
All prices are without VAT. Countries of the European Union will have to pay VAT in their own country. Please mention on your order the VAT (TVA ; MWS ; BTW ; IVA ; MOMS)-number of your company or institute. Our VAT no. is NL813037761B01. Without a VAT tax number, we have to add 19% VAT to orders shipped within the European Community.

3. If my country is part of the European Union, do I still have to pay VAT (TVA; MWS; BTW; IVA; MOMS)?
Invoices to countries of the European Union do not include any foreign taxes or INSTRASTAT. Total sales per European V.A.T. number is declared at the Dutch tax office every month, following the legal normative of the European Union. Customers within the European Union will be charged VAT tax by the tax office in their own country. Please mention on your order the VAT (TVA; MWS; BTW; IVA; MOMS) number of your company or institute.

4. Is it possible to obtain a SALSA MLPA sample kit?
No, unfortunately we do not have any test kits available. Every MLPA kit contains reagents and probemixes for 100 reactions. However, it is possible to order SALSA MLPA mixed kits, with up to 3 different probemixes (minimally 20 reactions each). Please note that each P-number is one probemix: e.g. the SALSA MLPA kit P034/P035 DMD consist of TWO probemixes.

5. Is it possible to order a part of a probemix / just one or more probes?
No, it is not possible to order a subset of probes from a certain probemix. Since all probes are already included in a single mix, one can only order the probe mix described in the product description. Nevertheless, it is possible to request for the design of a new probemix in which (some) existing probes are used. MRC-Holland may grant such a request if such a new probemix would be of interest to a wider audience.

6. Can I order a probemix for less than 100 reactions?
Every MLPA kit contains reagents and probemixes for 100 reactions. However, it is possible to order SALSA MLPA mixed kits, with up to 3 different probemixes (minimally 20 reactions each). 8. Is it possible to order custom-designed probes?
The number of requests for new MLPA probes is high. At this moment we are therefore only able to prepare "custom-designed probe sets" when these are expected to be of interest for more laboratories in the future. All costs for development of new probes are paid by MRC-Holland. Preparation of MLPA probes is time consuming and therefore expensive (>EUR 800 / probe). For each probe, a M13 clone has to be prepared, single stranded DNA from each clone is made and digested by expensive enzymes, etc.
We receive requests for the development of new probe mixes (or probes) each week. In case development seems worthwhile, one of our employees starts designing the requested probes or probe mix, usually in collaboration with the requesting lab(s). The lab usually sends us information about the presence of polymorphisms or common point mutations in the gene(s) involved, and about other genes which may cause a similar phenotype when defect. New probemixes are tested first at MRC-Holland and than offered to the requesting lab. Once the probemix seems to be useful for the application in question, the description of the kit is added to our website. In some cases, probemixes can be offered exclusively to a lab until their work has been accepted for publication ("new" genes). Please note that at least 50 different MLPA kits are available or in preparation, but not yet mentioned on our website.
In case new MLPA probes are required for research purposes only, we recommend designing synthetic MLPA probes using the information on the support section of our website. Synthetic probes consist of two synthetic oligonucleotides that can be ordered from your own oligo supplier. MLPA Reagents (without probemix) can be ordered from MRC-Holland (EK1, EK5 kits). The advantage of synthetic MLPA probes is that the probemix will be available much sooner. The disadvantage is a lower number of probes in one probe mix and perhaps a higher variability of synthetic probes as compared to our M13-derived probes.

Next to MLPA, we have been providing DNA molecular weight markers, restriction enzymes and DNA modifying enzymes (Ligases, Polymerases) to several major distributors since 1985. These items are usually only available only in bulk amounts (>EUR 1000 / item). Please enquire at info@mlpa.com..

10. Are there any other licenses required for performing an MLPA reaction?
This will vary from country to country. We do not expect problems for research purposes. The legal consequences of "patented genes" and "patented mutations" are not yet clear. Patent applications have been filed by MRC-Holland in various countries on certain aspects of the MLPA technique. For commercial applications, please contact MRC-Holland.

11. Do I have to pay in advance?
No. At the time of shipment, an invoice is sent by ordinary mail which is due approximately 30 days after shipment of the MLPA kits. Payment can be done by bank transfer (preferred) or by cheque; no credit cards.
